Conversely, if it is a smaller structure, such as virions and spores, a smaller lysis medium can be selected.

A blunt cleavage medium such as a sphere relies mainly on collision forces to lyse the sample; while a sharp cleavage medium relies primarily on shear forces to lyse the sample. Shear force, like a double-edged sword, is more conducive to breaking difficult samples, but it can also destroy RNA and some easily degraded proteins. Therefore, for DNA, stable proteins, polysaccharide structures, and small molecular substances, shear force is still very practical.

Hardness and density

The composition of the cracking medium determines two very important parameters: hardness and density. The hardness of the lysis medium must be higher than the hardness of the sample. Therefore, MP Bio's lysis medium can meet the requirements, so density is an important parameter. The higher the density, the better the grinding effect on refractory samples such as spores. In addition, the density of the cleavage media particles needs to be greater than that of the ground solution, otherwise the lysis medium floats on the surface and does not function as a cleavage or homogenization.
