Breast cancer can be treated by skin treatment to effectively absorb the drug

If a drug can prevent and treat breast cancer, but it has serious side effects, it can be effectively absorbed by the skin by a new topical drug delivery system.

Endoxifen, the active metabolite of tamoxifen, is one of the most commonly used hormones for the treatment of breast cancer and has also been shown to prevent breast cancer.

Researchers at the University of Illinois at Chicago collaborated with clinicians at Northwestern University to develop a chemical capsule for Endoxifen. Studies have shown that this chemical balloon technique is better than other techniques that help drugs pass through the skin barrier. The study was published in the recently published Prospective Functional Materials magazine.

"Endoxifen administration directly through the breast may reduce the number of cases of mastectomy and reduce the adverse effects of oral Endoxifen," said Seungpyo Hong, associate professor of medicine and bioengineering at the University of Illinois at Chicago.

Tamoxifen interferes with certain activities of estrogen and mimics other estrogen effects for the treatment of certain breast and ovarian cancers. According to reports, Endoxifen is an activated form of tamoxifen, which must pass through multiple layers of skin disorders to reach the target site that can prevent breast cancer. Endoxifen does not cross the skin barrier if there is no penetration enhancer.

The bladder invented by the researchers is a dendritic polymer micelle, which is actually a special bubble that wraps Endoxifen. They used hairless mice and human skin to test the ability of the balloon to cross the skin barrier and compare it to hydroxy tamoxifen without balloon encapsulation.

The results show that dendritic micelles can carry enough drugs to enhance skin permeability and control the release of hydroxy tamoxifen. Moreover, the efficacy was not reduced compared to free hydroxy tamoxifen.

According to Hong, chemical penetrants (such as alcohol or detergents) are small molecules that make the skin more permeable and may cause skin irritation.

Dendritic micelles deliver drugs in a more controlled manner, albeit at a slower rate than drugs delivered by cationic liposomes.

"Considering that the target audience is those who are likely to develop breast cancer, a slow drug delivery system may reduce the frequency of dosing and significantly increase patient compliance," Hong said.
